Intermediate - require GUI advice

Hi, I've used Python for data analysis scripts (throwing stuff together with NumPy and matplotlib) but am completely lost when it comes to user interfaces. Not just which libraries are most approachable, but also how to think about the problem of creating a UI in the first place.
